Spectralon® Fluorescence Standards Overview
Labsphere Spectralon® fluorescence standards are stable standards of diffuse fluorescence that are used for calibrating spectrofluorometers and fluorescence measurement instruments in general. Compared with glossy ceramic standards, Spectralon fluorescence standards provide a stable, consistent reference regardless of the instrument’s viewing or illumination geometry. Fluorescence materials aid in the development of optically brightened materials such as paper and textiles and are widely used in the cosmetic industry. Unlike disposable paper fluorescence standards, Labsphere fluorescence pucks provide highly stable, reproducible fluorescence reflectance over a long period of time in varying conditions.
Spectralon provides the ideal matrix for inorganic fluors which are photochemically stable when compared to their organic counterparts. The stability of the inorganic fluors, when combined with the durability of Spectralon, result in rugged, long-lasting fluorescence materials for both field and laboratory use. Labsphere’s fluorescence pucks are sold individually, in sets of three identical fluors, or as a set of the five fluors described below. Each puck is housed in a durable Delrin holder with a protective cover.
Calibration
Spectralon fluorescence standards are supplied uncalibrated. As such, they are not used to impart an absolute calibration onto the fluorescence measurement instrument, rather their utility is to serve as a stable baseline reference when comparing the fluorescence performance of sample materials. Typical spectral absorption and spectral emission data is provided for each standard type.

Standards are available individually and in sets of five in both 1.25 and 2.00 inch diameters, listed below. There are five fluorescent standard types to choose from, each with a characteristic excitation and emission spectra: blue/white (320nm excitation peak); blue/white (340nm excitation peak); blue; green; and orange.

Spectralon fluorescence standards are used whenever you need to baseline reference or validate the performance of your spectrofluorometer. Spectralon exhibits diffuse reflectance, often termed Lambertian. But what is a Lambertian reflector? Imagine a material which reflects light equally in all directions. It appears equally bright to an observer, regardless of the angle from which they view it. A material that exhibits Lambertian reflectance is one which is a perfectly matte, or diffusely reflecting. In physics, we say that the material obeys Lambert’s Cosine Law, named after Johann Lambert, who described the concept of a perfect diffuse reflector in his 1760 book “Photometria”.
In purely practical terms, by using a diffusely reflecting standard rather than a glossy plastic or ceramic material to calibrate your instrument gives you more consistent results, and results that can be compared between different instruments and between different laboratories. In comparison, a glossy calibration standard will impart a calibration that will depend upon the exact measurement geometry used in the particular instrument.
Spectralon fluorescence standards are used in the biomedical, pharmaceutical and textile industries as well as in the testing optical brightening agents, for example of „bright white“ materials in the paper industry.
